About the Book

This publication concentrates on the regulatory aspects of thrombin generation. Thrombin generation is an important process which is central to haemostasis and thrombosis. This unique biochemical system involves the proteolytic activation and inactivation of enzymes together with reactions at interfaces and non-enzymatic protein co-factors. The importance of the understanding of the influences of antithrombotic drugs on the mechanism cannot be overestimated, given the fact that some form of thrombosis (coronary infaction, stroke, pulmonary embolisms) accounts for almost half of the deaths occurring in the Western population. Both of biochemical and medical interest, the publication provides an overview of the role of feedback activation and inhibition as well as issues involved in therapeutic intervention.

Table of Contents:
Mechanisms in coagulation, H.C. Hemker and H. Kessels; simulation model for thrombin generation in plasma, G.M. Willems et al; interaction of feedback control and product inhibition in the activation of factor X by factors IX and VIII, J. Jesty; extrinsic pathway inhibitor - the key to feedback control of blood coagulation initiated by tissue thromboplastin, P.M. Sandset; modulation of the enzymatic activity of alpha-thrombin by polyanions - consequences on intrinsic activation of factor V and VIII, F.A. Ofosu; linearized model for the initiation of factor Va and thrombin generation, G.M. Willems; heparin requires both antithrombin and extrinsic pathway inhibitor for its anticoagulant effect in human blood, U. Abildgaard et al; determination of the levels of unfractionated and low-molecular-weight heparins in plasma - their effect on thrombin-mediated feedback reactions in vivo - preliminary results after subcutaneous injection, H.C. Hemker, et al.
