Abstract: Abstract of the thesis entitled Studies on effects of Coptis extract and Berberine against Carbon tetrachloride-induced liver damage in rats Submitted by YE Xingshen For the degree of Master of philosophy at the University of Hong Kong in August 2007 Coptidis Rhizoma (CR, Huanglian in Chinese), a famous Chinese herb listed in Chinese pharmacopoeia, has been extensively studied and is known to exhibit multiple pharmacological actions including antibacterial, antiviral, antiallergic, anti- inflammatory, anti-cholinergic, choleretic, antineoplastic, hypotensive, hypoglycemic and lowering-cholesterol effects etc.. CR has been used for two thousands years by Traditional Chinese Medicine (TCM) physicians to treat syndromes incurred by damp-heat, fire or toxicity in terms of TCM which can be conceived as inflammatory diseases nowadays. Although some experiments showed that CR only had preventive effect to Carbon tetrachloride-induced liver damage in rats, in fact CR was used for clinical purpose daily for treating chole-liver diseases, such as acute and chronic cholecystitis and hepatitis. The aim of this study was to investigate the effects of Coptidis and Berberine on damage of liver caused by carbon tetrachloride in rats. Rats were intraperitoneally injected with CCl and control ones with the same dose of olive oil. CR, suspended in distilled water at a concentration of 400, 600 or 800mg/kg, was orally administered to rats by gavage 6 hours after CCl treatment. The rats were sacrificed under ether anesthesia 24 hours after CCl treatment. At the same time blood was collected from the vena cave inferior and the livers were isolated. Serum enzymes including GPT, GOT and SOD, and histological study were estimated in all study groups. In other test, berberine was suspended in distilled water at a concentration of 80,120,160mg/kg, was orally administered to rats by gavage 6 hours after CCl treatment. One group was orally administered by berberine (120mg/kg) twice daily for 2 days before CCl treatment. After 24 hours of experiments, the rats, blood, livers and related parameter were observed same as the CR extract treatment. The results showed that CCl caused a marked rise in serum levels of GPT and GOT, 4 whereas SOD levels were decreased in rats. CR extract and berberine were administered to rats before and after the treatment with CCl which demonstrated significant effects on decreasing GPT, GOT, increasing SOD, and improving liver histological changes compared with the control group. The present results revealed that CR and berberine were effective in the protection of CCl4-induced liver injury in rats which was not only preventive, but also curative. As a possible mechanism, CR and berberine could scavenge oxidative-free radicals, inhibit lipid peroxidation and then alleviate liver injury. On the other hand, the model of CCl -induced hepatic injury in the rat simulates many of the features of acute hepatitis induced by toxicants and virals; hence our results could partially explicated therapeutic principle for CR in TCM clinical application and it is suggested that CR and berberine could be used as a potential new drug for acute liver injury.
