Abstract: Abstract of thesis entitled MOLECULAR AND FUNCTIONAL CHARACTERIZATION OF THE PROLACTIN RECEPTOR, PROLACTIN-RELEASING PEPTIDE RECEPTOR, AND GROWTH HORMONE-RELEASING HORMONE RECEPTOR GENES IN CHICKEN Submitted by WANG YING For the degree of Doctor of Philosophy at The University of Hong Kong in September 2007 PRL and GH are principally synthesized and secreted in the pituitary, and they are regulated by hypothalamic hormones, including prolactin-releasing peptide (PrRP) and growth hormone-releasing hormone (GHRH) through binding and activating their receptors, PrRP receptor (PrRPR) and GHRH receptor (GHRHR) respectively. However, chicken PrRPR and GHRHR have not yet been cloned and their functions remain largely unknown. In this study, chicken PrRPR (cPrRPR) was cloned from pituitary. Human PrRP 31 activates cAMP accumulation in cPrRPR-transfected cells in a dose-dependent manner, suggesting that cPrRPR is a receptor for PrRP. Expression of cPrRPR gene was found to be predominantly expressed in pituitary using RT-PCR, and its expression was found to correlate with cPRL expression during embryonic pituitary development, suggesting that cPrRPR may be involved in lactotroph differentiation. A full length cDNA of chicken GHRHR (cGHRHR) was isolated from pituitary. Synthetic chicken GHRH (cGHRH / ) activates cGHRHR with high affinity 1-31 1-47 (EC In addition, two novel cGHRHR transcript variants (cGHRHR-v1 and cGHRHR-v2) were isolated from pituitary. cGHRHR-v1 has a deletion of 36 amino acid residues 56 (50-85) including an aspartate residue (Asp ), which is conserved in all members of B-I GPCR superfamily. cGHRHR-v2 is a carboxy-terminal truncated transcript. Both GHRH and PACAP could activate cGHRHR-v1 at high dosages to induce cAMP accumulation after agonist treatment, whereas cGHRHR-v2 failed to respond. 56 Strikingly, site-directed mutants at Asp (Asp→Ala, or Asp→Gly) of the native cGHRHR reduced potencies (about 3 10-fold less potent) after agonist treatment. These results suggest that the residues (50-85) of cGHRHR, including highly 56 conserved Asp, may play an important role in the high affinity ligand-binding, instead of determining ligand-binding selectivity. PRL exerts multiple functions through first activating its membrane receptor, PRLR. At least 8 types of cPRLR transcript variants were identified and they can be divided into two groups (V1 and V2) according to their alternative usage of exon 1. Interesting, V1 transcript exhibits tissue-specific expression patterns and its expression in intestine is developmentally regulated during embryogenesis.
