Abstract: Abstract of thesis entitled PROTEOMIC ANALYSIS OF THE ANTI-INFLAMMATORY EFFECT OF TWO CHINESE MEDICINAL HERBS: DANSHEN AND YUNZHI Submitted by LIU Suk Yin Karen for the Degree of Master of Philosophy at The University of Hong Kong in June 2006 There are increasing evidence supporting the anti-inflammatory potentials of the two Chinese medicinal herbs, Danshen and Yunzhi. However, the mechanistic of actions have not yet been fully explored. In an attempt to establish the anti- inflammatory mechanism(s) of Danshen and Polysaccharopeptide (PSP) derived from Yunzhi, an in vitro Lipopolysaccaride (LPS)-induced inflammation in RAW 267.4 macrophage model was employed in this study. Western blot and molecular biology techniques were used to investigate the effect of Danshen and PSP on the inflammatory pathways induced by LPS by measuring the expression of the Toll-like 2 (TLR-2) and Toll-like 4 (TLR-4) receptors; the pro-inflammatory proteins cyclooxygenase-2 (COX-2), microsomal prostaglandins E (mPGE ); the inducible nitric oxide synthase (iNOS); and proteins 2 2 involved in signaling pathway such as the transcriptional factor nuclear factor-kappa B (NF-κB) and p38 mitogen-activated protein (MAP) kinases. In addition, two dimensional electrophoresis (2-DE) proteomic analysis of proteins change was employed to identify the disease-relevant biomarkers and to explore the effect of Danshen and PSP on the disease-relevant biomarkers for alleviating the inflammatory process induced by LPS. The results show that Danshen and PSP exhibited significant inhibition but by different extent on the TLR-2 and TLR-4, the COX-2, iNOS, mPGE protein expression. Similar down-regulation effect of PSP and Danshen on NF-κB and p38 MAPK pathway was observed. Proteomics analysis revealed the dynamic change of proteomes over the 24 hours duration of investigation. The proteins affected by the different duration of LPS exposure are the Hyaluronan receptor, STAT 1 protein and nitric oxide sensitive guanylyl cyclase beta 2 subunit. The following proteins DNAJC 7, Dynamin 2, Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and Prohibitin were up-regulated by the treatment of PSP and Danshen. Other cellular proteins such as the Calpain 7; Hyaluronan mediated molity receptor, Synembryn and Tropomyosin 3 were, however, greatly suppressed with Danshen and PSP treatment. In conclusion, the molecular approach applied in this study provides new insights into the anti-inflammatory molecular pathways of Danshen and PSP. The high- resolution of the 2DE and protein identification technique also allow rapid discovery of new potential candidate proteins as biomarkers during Gram-negative bacterial infection.
