About the Book

Somatic Hypermutation (SHM) is a process by which point mutations are introduced in the variable regions of Immunoglobulin (Ig) genes in activated B cells. The mutations alter the binding affinity of the resulting Ig proteins so that some of the immunoglobulins will be better able to bind the activating pathogen. SHM is dependent on the enzyme Activation Induced Cytidine Deaminase (AID), which deaminates cytidine in the DNA. Until recently it had been assumed that SHM is targeted to Ig genes. However, a study by Man Liu determined that more than 25% of the expressed genes in activated B cells are mutated in an AID dependent manner (1). The list of mutated genes includes the oncogenes Bcl6, Pim1, and cMyc.I hypothesized that the mutating non-Ig genes may mutate simply because they are near Ig genes; their mutation might be collateral damage incurred during SHM. To investigate whether there is a correlation between proximity to Ig genes and mutability of non-Ig genes, I used 3D-Fluorescence in situ Hybridization (FISH) to measure the distance from the genes of interest to either IgH or Iglambda. No correlation between mutability and proximity was observed. Furthermore, cMyc was not found to be closer to IgH than are other mutating genes, indicating that its increased frequency of translocation with IgH is probably not due to an abnormally close association of IgH and cMyc in GC B cells.In an attempt to better understand how the position of a gene affects its mutation, I also made mice transgenic for human MYC and analyzed the expression and mutation of the transgenes. The human MYC transgene was not deaminated by AID in either of two locations. One of the MYC transgenes, huMyc24, was expressed only at very low levels in Peyer's patch GC B cells and this lack of expression is the most likely cause of its lack of mutation. The other transgene, however, was expressed in all cell types analyzed. Its lack of mutation is probably due to a difference between the human and mouse versions of the gene or a change in nuclear localization and local chromatin context.
