Small GTP-binding proteins are best known for their ubiquitous contribution to signaling cascades in eukaryotic cells. Except for the pioneering work on G proteins involved in translation, the physiological functions of many prokaryotic G proteins remain elusive and their biochemical characterization is largely incomplete. Among the few exceptions is the GTP-binding domain that is embedded in the cytosolic, N-terminal domain (NFeoB) of the polytopic membrane protein FeoB. The FeoB family of G proteins is involved with high affinity Fe(II)-uptake in prokaryotes, which is required for the virulence of pathogens such as E. coli, S. typhimurium and H. pylori. FeoB belongs to the TRAFAC (after translation factors) class of P-loop NTPases and has a unique molecular design by combining at least 7 transmembrane helices with a GTP-binding domain. In vitro, NFeoB binds GDP and GTP with moderate affinity and, similar to other prokaryotic G proteins, has rapid exchange rate constants for either nucleotide. The cytosolic regions of FeoB contain a domain (IIFeoB) that bridges the N-terminal GTP-binding domain with the C-terminal membrane domain and functions as a G protein regulator by specifically affecting GDP affinity. The isolated IIFeoB domain directly interacts with the GTP-binding domain switch regions that are required for function to decrease the dissociation rate of GDP binding. The structure of the apo form of NFeoB from Pyrococcus furiosus was determined at 2.7 A. IIFeoB is comprised of a bundle of five alpha helices, which resembles other effectors with helical scaffolds that regulate P-type NTPases. The discovery of this prokaryotic GDP dissociation inhibitor-like domain suggests that Fe(II)-uptake in bacteria involves a G protein regulatory pathway reminiscent of signaling mechanisms found in higher order organisms. Taken together, these findings raise the question whether FeoB-dependent Fe(II)-uptake is an archetypal predecessor of G protein-coupled membrane processes that are ubiquitously found in higher organisms.
