Genetic, biochemical, and neuropathological studies support the idea that cerebral elevation and accumulation of amyloid beta-peptide (Abeta) are early and necessary steps in the pathogenesis of Alzheimer's disease (AD). Abeta is produced by sequential proteolytic cleavage of beta-amyloid precursor protein (APP) by a set of membrane-bound proteases termed beta- and gamma-secretases. BACE1 was identified as the major proteolytic activity of beta-secretase that mediates the first cleavage of APP. Because BACE1 knockout mice are viable, fertile, and do not produce Abeta, BACE1 has become a popular target for AD therapeutic intervention. We developed a novel cell-based assay that can directly monitor BACE1-mediated cleavage of APP in intact neuronal cells. Unlike cell-free BACE1 assays, we have the potential to discover indirect small molecule modulators of BACE1 that are effective in intact cells in addition to direct BACE1 inhibitors. In collaboration with the Laboratory for Drug Discovery in Neurodegeneration (LDDN), we conducted a high-throughput chemical genetic screen with a 140,000-compound library and identified 5 molecules capable of reducing BACE1-mediated cleavage of APP in the screening cell line. Structure-activity relationship studies and subsequent characterization in primary neurons and in Tg2576 and PS-APP Alzheimer model mice revealed one potent and blood-brain barrier-penetrant chemotype that reduced BACE1-mediated cleavage in vitro and in vivo without inhibiting BACE1 directly. Discovery of an indirect inhibitor class may lead to identification of a novel BACE1-regulating protein, and may offer a novel therapeutic target for the treatment of Alzheimer's disease.
