Injuries to the corticospinal (CS) system, such as after stroke or spinal cord injury, usually are incomplete leaving spared CS connections between the primary motor cortex and termination sites throughout the spinal cord. There is increased interest to utilize these spared connections to promote recovery of cortically mediated motor control. In particular ipsilateral CS fibers spared after injury might be able to restore control of spinal motor circuits previously innervated by the damaged contralateral CS tract (CST). Since these connections comprise less than 10% of all CS fibers, augmenting their number and strength may improve CS mediated function after injury. In this dissertation I examined whether selectively augmenting spared CS terminations with activity---a crucial factor in CS development---can strengthen their connections to spinal motor circuits and, in turn, promote recovery from incomplete CS injury. I hypothesized that augmenting neural activity would improve spared CS function and promote recovery of motor behaviors dependent on the CS system. I found that selective stimulation of the intact CST increased the connectivity of spared ipsilateral connections and improved recovery in a test of visually-guided locomotion. These results suggest an important role of the ipsilateral CST in restoring control after injury, yet they also raised questions about role of the ipsilateral CST. In a series of cortical mapping studies I found an extensive ipsilateral forelimb motor representation that requires a complex set of interactions from the other cortical hemisphere to elicit ipsilateral responses. While these findings imply a greater role for ipsilateral CS fibers in normal motor function, and may indicate additional sites where selective stimulation could promote ipsilateral CS connectivity.
