About the Book

Every individual is unique due in part to natural sequence variations in our DNA. The same degree of polymorphism exists when the genomes of two wild isolates of C. elegans, a common microscopic soil nematode, are compared. To explore the impact of these variations, I generated a collection of 800 recombinant inbred lines (RILs) by interbreeding two C. elegans wild isolates. Two robust and healthy strains, Bristol (N2) and Hawaiian (CB4856) isolates, have been evolving in relative isolation for thousands of years. Over time, N2 and CB4856 have diverged to the point where the strains are roughly as distinct as any two humans might be from each other. Each resultant RIL was genotyped with single nucleotide polymorphism (SNP) markers and as expected, has a unique, mosaic genome contributed by the two wild isolates. From lifespan data measured in a subset of RILs, I detected five novel longevity quantitative trait loci (QTLs) using linkage analysis. Additional QTLs were predicted in daf-16 sensitized background. These putative QTLs may function independent of DAF-16, a forkhead transcription factor (FOXO) and a known regulator of aging. Identification of the gene(s) underlying each QTL is possible with genetic approaches documented in this thesis. Elucidating the functions of these genes can lead to further understanding the mechanism of aging. Furthermore, since SNP-map of all RILs is available, any researcher can utilize the strains to study their favorite biological processes in its most natural states.
