About the Book

Steroid hormones are unique compounds in that they are active at the interface of peripheral endocrine events and neural mechanisms. Thus their effects present an important peripheral signalling system to alter brain function. This volume presents state-of-the-art and classical techniques for the study of steroid hormones and their receptors and their effects and actions. Comprehensive protocols are included for the study of: steroid kinetics and metabolism; steroid receptors; molecular and cellular effects of steroids; and steroid effects on integrated systems.

Table of Contents:
Part 1 Steroid kinetics and metabolism: steroid hormone transport through the blood-brain barrier - methods and concepts, W.M. Pardridge; enhanced delivery of steroids to the brain using a redox-based chemical delivery system, J.W. Simpkins and N. Bodor; neurosteroids - biosyntheisis and function, P. Robel and E.-E. Baulieu; cytochrome P450 enzymes in brain, M. Warner et al; methods for estimating 11b-hydroxysteroid dehydrogenase activity, Z. Krozowski. Part 2 Steroid receptors: steroid hormone binding to intracellular receptors - "in vitro" and "in vivo" studies, A.D. Van Harst and T.F. Szuran; steroid hormone binding to membrane receptors, M. Orchinik and T.F. Murray; "in vitro" autoradiography for steroid receptors, N.J. MacLusky et al; immunocytochemical studies on glucocorticoid receptor, A. Cintra et al; confocal scanning laser microscopy of steroid receptors in brain, B. van Steensel et al; ultrastructural aspects of steroid receptor localization - an immunocytochemical perspective, Z. Liposits and I. Kallo; hybridization studies of adrenocorticosteroid receptors in central nervous system, J.P. Herman et al; detection and characterization of epalon receptors - novel recognition sites for neuroactive steroids that modulate the GABAA receptor complex, L.D. McCauley and K.W. Gee; mutation analysis of steroid hormone receptors, M. Karl et al. Part 3 Molecular effects of steroids: protein-DNA binding assay for analysis of steroid-sensitive neurons in mammalian brain, Y.-S. Zhu and D.W. Pfaff; gene transfection studies using recombinant steroid receptors, K. Damm; regulation of neuropeptide genes - determination of responsiveness to steroids and identification of receptors in brain nuclei, J.J. Cox et al; cloning of steroid-responsive mRNAs by differential hybridization, N.R. Nichols et al; molecular correlates of corticosterone action in hippocampal subregions, S.M. Nair and J.H. Eberwine; glucocorticoid-determined protein synthesis, L.A. Dokas; use of antisense oligodeoxynucleotides to block gene expression in the central nervous system, M.M. McCarthy. Part 4 Cellular effects of steroids: steroid regulation of neuronotrophic activity - primary microcultures of midbrain raphe and hippocampus, E.C. Azmitia and X.-P. Hou; central glucocorticoid receptors and neuronal plasticity, K. Fuxe et al; steroid action on neuronal structure, C.S. Woolley and E. Gould; electron microscopic double and triple labelling immunocytochemistry in elucidation of synaptological interactions between ovarian steroid-sensitive neurons and circuits, C. Leranth et al; gene-mediated steroid control of neuron activity, M. Joels; electrophysiological studies of neurosteroid modulation of [gamma]-aminobutyric acid type a receptor, C. Hill-Venning et al; "in vitro" approaches to studying glucocorticoid effects on gene expression in neurons and glia, M.C. Bohn. Part 5 Steroid effects on integrated systems. (Part contents).
